MesenCult™-ACF Plus Umbilical Cord Culture Kit (ST-100-0234) は、ヒト臍帯組織から間葉系間質細胞 (間葉系幹細胞、MSCとも呼ばれる) を誘導および増殖する動物性成分フリー (ACF) の培地です。本品をMesenCult™-ACF Plus Medium 完全培地と併用することで、臍帯由来MSC (UC-MSC) を長期増殖できます。
完全なACF培養系を実現するには、MesenCult™-ACF Plus Umbilical Cord Mediumを、Animal Component-Free Cell Attachment Substrate および Animal Component-Free Cell Dissociation Kit (ST-05426) と組み合わせて使用する必要があります。これらは、その成分がスクリーニングされており、本品および MesenCult™-ACF Plus Medium での培養時に最適な細胞接着が起こることを検証済みです。
MesenCult™培地で培養したヒトMSCを、ACF条件で最適に凍結保存するには、MesenCult™-ACF Freezing Medium（ST-05490）の使用を推奨いたします。
注：MesenCult™-ACF Plus Umbilical Cord Medium および MesenCult™-ACF Plus Medium 完全培地の調製時には、L-Glutamine (ST-07100) を添加してください。

Protocol for deriving and expanding umbilical cord-derived mesenchymal stromal cells (UC-MSCs) with the MesenCult™-ACF Plus Umbilical Cord Culture Kit.
UC-MSCs are derived in 14 days using MesenCult™-ACF Plus Medium supplemented with the MesenCult™-ACF Plus Umbilical Cord Derivation 500X Supplement. After Day 14, cells can be maintained in complete MesenCult™-ACF Plus Medium. Representative images at Day 6, 14, and 21 are included for reference.

Compared to MesenCult™-ACF Plus (MACFP) medium, the isolation yield, per gram umbilical cord tissue, of UC-MSCs in MesenCult™-ACF Plus Umbilical Cord (MACFP-UC) Culture Kit is 65% higher. The addition of 2.5% human AB serum during isolation further increases the yield to over 4 times more than with MesenCult™-ACF Plus alone.

(B) The morphology of UC-MSCs from two donors remains MSC-like following 21 passages in MesenCult™-ACF Plus Medium.
